Here's a list of highlights that I copypasta'd from reddit:

Canterlot is based off of Minas Tirith
Parents of Celestia, Luna were intended to be explored, but because of lack of time in first season were not
Scootaloo was created as a character who could not fly, and whose central confict arises from her idolization of RD's ability to fly
Ditzy was a concept for a character, but she liked Derpy and wanted to run with her instead as a gag ( like in Feeling Pinkie Keen with the Anvil), meaning Ditzy canonically never appeared on screen
She concieved the Changlings as an enemy
Idea for episode: RD and AJ find a pony raised by deer and who believed he was a deer (branches tied to head)
She is working on new show "Wander Over Yonder" with husband for Disney

-0:32 - Canterlot was intended to be based off of Lord of the Rings ("Gondor for little girls")
-0:48 - Mane 6 & Princesses' parents - never got to flesh any of them out at all besides what we have
-1:24 - Twilight inspired by Lauren's mother
-1:32 - Fluttershy inspired by some of Lauren's childhood moments
-1:50 - Dash inspired by Firefly (we knew this anyways)
-2:01 - Pinkie was intended to be a free spirit (I don't really see any point here, but :p inkieshrug: )
-2:24 - Lauren knew Tabitha was right for Rarity right away :vogue:
-2:44 - Lauren had ~3 Scootaloo episodes in mind focused on being technically handicapped - the character was designed to never be able to fly and struggle with her dreams of being like R.D. and reality
-4:10 - Lauren already had an idea for a BG character that would be a clumsy troublemaker, named her Ditzy Doo, was going to create a fully new character for Ditzy but when heard about Derpy phenomenon went with that, WWU Ditzy mention happened before they found out, FPK cameo happened after
-5:13 - Lauren intended Derpy to be side gag character occasionally causing problems w/ no voice
-5:27 - Lauren came up with concept of changelings as the enemy, did not design them visually at all, based off of some mythology
-6:14 - Episode idea that never got past idea stage: R.D. / A.J. have some type of adventure ending up in White Tail Wood, encounter deer family w/ pony w/ branches tied to his head (raised by deer after abandonment), sentience of deer unspecified, Dash wants to tell pony that he is a pony, AJ says it isn't their place, resolution was never thought of (another RD/AJ ep in White Tail Woods? Did FWF replace this?)
-7:30 - Lauren thinks difference between PPG fandom and this fandom is the state of the Internet at the two times

Yeah, I'm pretty jealous.

By the by, my sister got me a Pony poster the other day as an early Christmas/Birthday/Next Christmas present; she knew someone who was interning at DHX. It looks pretty cool; it has the pony OCs for the director (Thiessen), music guy (Ingram?), Tara Strong, Sibsy (Wildfire) on centre stage. It's signed by Wootie, some character designers, and some other signatures I can't make out.

I don't really know what to do with it.
